Application
 
- Transfer short oligonucleotides (e.g. DNA, RNA, siRNA, miRNA)
- Transfection of primary difficult-to-transfect cells, including human embryonic stem (ES) cells, human fibroblast cells
- In vivo gene delivery zygote, egg and sperm (e.g. zebrafish, xenopus, porcine etc)
- Development study
- Gene delivery study according time lap
- HTS study (high-throughput)
 
Protocol for Treatment of the VisuFect® 100 for Zygote
 
A. Prepare VisuFect® 100 stock (100pmole/ul).
B. Drop VisuFect® 100 stock (to be final conc. 1 pmole/ul)* into culture medium which contains Zygote or Sperm.
C. Mix plate gently.
D. Incubate for 12-16 hrs at CO2 incubator.
E. Wash for 3 times and change to fresh culture medium.
F. Visualize transfected cell with a confocal microscopy. (Ex.675nm / Em.694nm)
* It is necessary to optimize VisuFect® 100 concentration for customer’s experiments

VisuFect® 100 Gene Delivery Reagent


Description

 

VisuFect® 100 is made up of an organic compound and chemically for the uptake by cells without a transfection reagent. VisuFect® 100 can deliver oligonucleotides in which a user id interested such as DNA aptamers, siRNAs, miRNAs or antisense RNAs into cell without any transfection reagent.
Since VisuFect® 100 contains fluorophore, the researcher can easily monitor efficiency of transfection.
VisuFect® 100 is a customized product which made by customer’s own sequence. VisuFect® 100 reagent is ideal for transfection of various cells, including primary difficult-to-transfect cells zygote, egg and sperm.
Transfection can also be performed regardless of the presence or absence of serum.
VisuFect® 100 demonstrates superior transfection efficiency and minimal toxicity compared with the lipid-based or other polymer-based transfection reagents.

Figure 3. Low cytotoxicity of VisuFect® 100. A comparison of development rate of blastocyst between VisuFect® 100 treatment and microinjection using by same siRNA. It shows almost same development rate between control and VisuFect® 100 samples. However, in the case of microinjection, the development rate of blastocyst was reduced.
